Des O'Conner and Melanie Sykes have been axed by ITV as part of the channel's mass schedule revamp.
Their chat show, Today With Des And Mel, has proven consistently popular since its launch in January 2002.
However, new ITV daytime commissioner Liam Keelan believes that the show has run its course and is now looking for fresh ideas to win more viewers.
An ITV spokeswoman said: "Des and Mel have done a fantastic job and we're very grateful.
"The daytime schedule is being overhauled and we have some exciting new programmes lined up."
It is not yet known whether O'Connor, 74, and Sykes, 35, will be kept on by ITV but the former's spokesperson confirmed he will stay on TV.
"It's always been enormously successful," the spokesperson said. "Des has enjoyed every moment and he's looking forward to working on other projects."
